Preferential accumulation of meloxicam in inflamed synovial joints of dogs.

چکیده
MELOXICAM is a NSAID classified as a derivative of enolic acid, licensed to treat acute and chronic musculoskeletal disorders in the dog. Meloxicam achieves maximal plasma concentration 4.5 hours after oral administration, has a low volume of distribution (0.3 l/kg), is highly plasma protein bound (97 per cent) and has an elimination half-life of 24 hours in dogs (EMA 2011). Steady state pharmacokinetics are achieved within one day using the licensed dose rate in the dog (EMA 2011). Preferential accumulation and persistence of meloxicam in inflamed joints of both the rat and the human being have previously been documented (Busch and Engelhardt 1990, Lapicque and others 2000). The objective of this study was to investigate the concentration of meloxicam in the synovial fluid of inflamed joints versus non-inflamed joints in dogs at the lowest plasma concentration (ie, trough level), 24 hours postdosing.

Effect of Angiotensin II on Blood Flow in Acute and Chronically Inflamed Knee Joints of Rabbits: The Role of Nitric Oxide
Background: Angiotensin converting enzyme (ACE) upregulation in stromal cells of joints affected by rheumatoid arthritis may lead to higher tissue angiotensin II that is a vasoconstrictor and mitogen factor. To date, the role of angiotensin II on regulating blood flow in inflamed joints has not been studied. Methods: Acute and chronic joint inflammation was induced in rabbits by intra-articular...
متن کاملBorrelia burgdorferi migrates into joint capsules and causes an up-regulation of interleukin-8 in synovial membranes of dogs experimentally infected with ticks.
Twenty 6-week-old specific-pathogen-free beagles were infected with Borrelia burgdorferi by tick challenge, and five uninfected dogs served as controls. During the study, all dogs were monitored for infection, clinical signs, and antibody response against B. burgdorferi. During episodes of lameness or postmortem, synovial fluids from each dog were examined for volume, cell number, polymorphonuc...
متن کاملMichael Mason prize essay 2003. Why do leucocytes accumulate within chronically inflamed joints?
Chronic inflammation is characterized by the accumulation of leucocytes within tissues. In rheumatoid arthritis the inflammatory infiltrate shares many architectural features with lymphoid tissue. For example, CD4 T cells and B cells accumulate in perivascular lymphoid structures within synovial tissue. CD8 T cells and neutrophils are found predominantly within synovial fluid. What drives these...
متن کاملThe Effect of Intra-Articular Injection of Two Anti TNF Drugs on Histopathology and Effect of Local Infusion of these Drugs on Blood Flow of Chronically Inflamed Joints in Rabbit
Background & Aims: The anti TNF drugs are new line treatment for articular inflammatory diseases which are used systemically, but due to expensiveness and some systemic adverse effects have limited usage. The aims of this study were to investigate the effect of intra-articular (local) injection of Etanercept and Infliximab as two anti TNF drugs on histopathology and also due to the important ro...
متن کاملEvaluation of intra-articular and subcutaneous administration of meloxicam for postoperative analgesia following stifle surgery in dogs.
The objective of this study was to compare the efficacy of meloxicam when given by intra-articular (IA) and subcutaneous (SC) routes of administration for postoperative analgesia versus a placebo for dogs undergoing stifle surgery. Twenty-five dogs with cranial cruciate ligament rupture (CCLR) were randomly assigned to one of three treatment groups, each with nine dogs, before surgical repair o...
